Digital quantification of proteins and mRNA in single mammalian cells

Research concerning single-cell protein and mRNA counting, led by Professor Savas Tay, was recently published in Molecular Cell, a high-impact journal with an impact factor (IF) of 14.

Albayrak et al. combine digital Proximity Ligation Assay (PLA) and digital RT-PCR to conduct joint and ultra-sensitive quantification of protein and mRNA from single mammalian cells.

The developed workflow was used to investigate the expression of an endogenous gene, and revealed a low correlation between the two macromolecules. Finally, the collected data was used to parameterize a stochastic model of the central dogma.
